Optical imaging is a rapidly developing field of research aimed at non-invasively interrogating animals for disease progression, determining the effects of a drug on a particular pathology, assessing the pharmacokinetic behavior of a drug, or identifying molecular biomarkers of disease. One of the key components of molecular imaging is the development of specific, targeted imaging contrast agents to assess these biological processes. The development of robust fluorochrome-labeled optical agents is a process that is often underestimated in terms of its complexity. Although many studies describe the use of these agents, guidelines for their development and testing are not readily available. This review outlines some of the general principles that are important when developing and using fluorochrome-labeled optical contrast agents for oncology investigations in animals.

Alterations in glycosyl epitopes play key roles in tumor progression. Accurate and reliable means of variety identification are necessary to assess purity of seed supplies, to support claims relating to plant breeders’ rights and, in Canada, to provide quality assurances in the grain handling system. A single, multiplexed set of seven simple-sequence-repeat (SSR) markers was found to uniquely identify all 18 durum wheat varieties that have been developed in Canada and are currently, or were formerly, registered for commercial production. Significant features of this multiplexed set include an allele that is specific, within Canadian durum varieties, to those having high gluten strength, and redundancy that was included in an effort to increase the capacity to accommodate future varieties. In combination with a reasonably rapid individual-kernel DNA extraction protocol and automated allele calling, this marker system offers a higher resolution alternative to complement established protein-based variety identification methods.

This paper reviews a newly developed miniature dual-axes confocal microscope. It has been used for subsurface imaging of biological tissues with 5–7 u m resolution. Reflectance and fluorescence images were obtained with a laser source at 785 nm and the IRDye 800CW.

Photon penetration into living tissue is highly dependent on the absorption and scattering properties of tissue components. The near-infrared region of the spectrum offers certain advantages for photon penetration, and both organic and inorganic fluorescence contrast agents are now available for chemical conjugation to targeting molecules. This review focuses on those parameters that affect image signal and background during in vivo imaging with near-infrared light and exogenous contrast agents. Recent examples of in vivo near-infrared fluorescence imaging of animals and humans are presented, including imaging of normal and diseased vasculature, tissue perfusion, protease activity, hydroxyapatite and cancer.

In vertebrates, the development and integrity of the skeleton requires hydroxyapatite (HA) deposition by osteoblasts. HA deposition is also a marker of, or a participant in, processes as diverse as cancer and atherosclerosis. At present, sites of osteoblastic activity can only be imaged in vivo using -emitting radioisotopes. The scan times required are long, and the resultant radioscintigraphic images suffer from relatively low resolution. We have synthesized a near-infrared (NIR) fluorescent bisphosphonate derivative that exhibits rapid and specific binding to HA in vitro and in vivo. We demonstrate NIR light?based detection of osteoblastic activity in the living animal, and discuss how this technology can be used to study skeletal development, osteoblastic metastasis, coronary atherosclerosis, and other human diseases.
